Genome-wide identification and functional characterization of glutathione peroxidase genes in date palm (Phoenix dactylifera L.) under stress conditions

Glutathione peroxidase (GPX) (EC 1.11.1.9) is a group of antioxidant enzymes that catalyze the reduction of hydrogen peroxide (H2O2) and lipid peroxidase to water and lipid alcohols, respectively. GPX enzymes are indispensable in protecting the cell from reactive oxygen species (ROS) damage, stress signaling and for maintaining cellular redox balance under stress conditions. Date palm (Phoenix dactylifera L.) is an important fruit-producing tree in the Arabian Peninsula, which can tolerate extreme environmental conditions, including salinity and drought conditions. This tolerance trait may be attributed to the various antioxidant enzyme system of the plant. Therefore, in this study, we aim to understand the contribution of the GPX gene family to the antioxidant system present in date palms. Five putative PdGPX genes (PdGPX1–5) were mined from the date palm genome, computationally analyzed, and their expression profiles under salinity, oxidative and methylglyoxal stresses, were determined using qPCR. The identified PdGPX genes were then functionally characterized at the molecular level in Escherichia coli (BL21) and the yeast (Saccharomyces cerevisiae) knockout cell line (GPX3Δ). The results showed that the PdGPX gene family is conserved in the date palm genome and their expression was altered based on the stress used in the experiment and plant tissue. Furthermore, heterologous expression of the PdGPX genes enhanced the survival of the E. coli (BL21) under different stresses, including salinity (250 mM NaCl), toxicity (0.5 mM methylglyoxal (MG) and oxidative stress (5 mM H2O2). The putative PdGPX genes were also able to complement the loss in function of the yeast knockout lines (GPX3Δ) by enhancing their survival under H2O2 and MG stress. These findings indicated that the PdGPX family plays a vital role in the antioxidant activity, which may enhance the survival of the date palm under stress conditions.
