Effect of pre and perinatal exposure to Bisphenol-A on the development of atherosclerosis in adult offspring

Description

Bisphenol-A (BPA) is the commonest constituent in polycarbonate plastic production. Its widespread exposure in day-to-day life through the usage of plastic has imposed concern about its toxic effects on public health. BPA can cross the placenta and its exposure during the early stages of growth can accelerate adipogenesis in offspring and these effects seem to continue into adulthood. Evidence from an in vitro study suggests that BPA exposure alters the vascular endothelial cells functions and also increases the inflammation markers. In the present study, we intend to investigate the effects of pre and perinatal exposure of BPA on development of atherosclerosis in adult offspring. Pregnant apoe?/? mice will be treated with low (50 ?g/kg) and high (1250 ?g/kg) doses of BPA throughout the gestation and continued till weaning. Adult offspring of experimental mice, at the age of 14th week will be assessed for the development of atherosclerosis and structural changes in liver and kidney. After cervical dislocation, aorta and peripheral arteries will be collected and subjected to gross atherosclerotic lesions quantification, transmission electron microscopic examination and histological analysis. Kidney and liver tissues will be collected for the histopathological analysis. Results of the present study will reveal whether maternal BPA exposure can cause atherosclerosis in adult offspring. Findings of such positive association will caution public especially pregnant women about restricted use of plasticware in day-today-life.
